The second article is saying something like this:
The polymyxin acted to partially block the effect of stimulating a nerve. They tested this on a preparation where a particular nerve normally causes a muscle contraction. They found that polymyxin B reduced that muscle contraction. The effect was not on the nerve, because the nerve still released acetyl choline. Therefore it worked by blocking the muscle. They proved this by applying the polymyxin, and then squirting acetyl choline onto the muscle, thus bypassing the nerve. The muscle contraction was blocked even more effectively. They think that the mode of action is to make the muscle less sensitive to acetyl choline.
Acetylcholine is normally broken down by cholinesterases. If you block the cholinesterases, this antagonizes the effect of the polymyxin (because now the acetylcholine hangs around for longer).
The other articles seem to say similar things.
Does this make sense? It does say that polymyxin and other similar antibiotics can be muscle relaxants, which I had never heard of before, and sounds like very good news for the likes of us.
